Claudin 7 belongs to a family of 24 tight junction (TJ) membrane proteins which are central for the regulation of cell permeability and the maintenance of cell polarity. Recent antibody-based studies indicated that Claudin 7 is not only localized at the apical TJs but also have a strong basolateral membrane distribution in the epithelia of various tissues. Abnormal expression and mislocalization of Claudin 7 are frequently observed in epithelial-derived lung, breast, head, and neck cancers. Measuring the Claudin 7 levels is important for the detection of different types of cancers and is also an important target for therapeutic intervention.
AlphaLISA technology allows the detection of molecules of interest in a no-wash, highly sensitive, quantitative assay. In an AlphaLISA assay, a biotinylated anti-analyte antibody binds to the Streptavidin-coated Donor beads while another anti-analyte antibody is conjugated to AlphaLISA Acceptor beads. In the presence of the analyte, the beads come into close proximity. The excitation of the Donor beads causes the release of singlet oxygen molecules that triggers a cascade of energy transfer in the Acceptor beads, resulting in a sharp peak of light emission at 615 nm.
